Proteasome Substrate II, Z-LLE-AMC, is a fluorogenic substrate for the peptidylglutamyl-peptide hydrolyzing (caspase-like) activity of the proteasome. Z-LLE-AMC's Caspase-like activity can be quantified by fluorescent detection of free AMC (7-amino-4-methylcoumarin). AMC is excited at 340-360 nm and emits at 440-460 nm. Z-LLE-AMC can be stimulated in the presence of Mg2+ ions and rapidly inactivated by N-acetylimidazoleis. |
